In the Settings > Working Hours & Days Off. i have updated that all week days are working days, nevertheless the calendar of Appointments in wp-admin has Sat & Sun disabled, please look into this issue as mostly i do appointments from wp-admin
You need to adjust the work hours for each employee individually in employee modal in work hours. You have set general work hours and this will affect only new emplyoee once you create them but the employees that you already have have default settings mon-fri 09-17 so you need to set this in employee work hours and all should be fine.

Are you using the Admin wordpress role to set the appointment?
