With out host provider, pantheon.io, our site operates with a development and test tier that carry their own URLs and I was wondering if this would count towards a domain each. Some of our plugins ignore a pantheonsite.io URL or a URL that contains test- or dev- and I was wondering if wpDataTables does this.

Yes. With our licences, you get an unlimited number of sub-domains/sub-sites.
So, if you take a Basic licence, for example - and have multiple Sites on the same Domain, you will be able to activate the licence on all of them/as long as they are under the same domain.
