You can achieve this in 2 ways. For example, you can enter on your calendar (if you have integrated Google or Outlook calendar) that you have an appointment or that you are busy at a certain time on a certain day and that slot will not be available for booking in Amelia.

Another way is to create a special day in Amelia/employee/edit employee/special days and set there at what time and on which service the employee will work and that's it.
