I am sorry to disappoint you, but unfortunately something like this is not possible with the plugin's built-in features.

You cannot apply the coupon to an existing appointment, and you can't apply it from back-end. The coupon option is only for customers, so you cannot do anything with it from back-end. You could use it in your customer's stead if you book for them from the Incognito window.
