Do you mean on something like that https://datatables.net/examples/api/counter_columns.html (This column should not be sortable, and will change dynamically as the ordering and searching applied to the table is altered by the end user) or just normal index column that will be not updated dynamically?

Then it is possible. If you are creating Manual wpDataTable there will be always wdt_id column that will be primary key in the database table. Otherwise if you are creating table that is not Manual (Excel, CSV, Google Spreadsheet...) you can always create column like that in your data source file, and create wpDataTable from it.
